Ponatinib is an anticancer drug. It was approved by the FAD and entered into market in 2012 12 for the treatment of adults with chronic myeloid leukemia (CML), "Philadelphia chromosome positive" (Ph +) and acute lymphoblastic leukemia (ALL). It is mainly used for the treatment of patients who have failed to get any response from dasatinib or nilotinib therapy or are intolerant to dasatinib or nilotinib and patients who are not suitable for being subject to follow-up treatment of imatinib. It can also be used for treating patients with a gene mutation ("T315I mutation ') which makes the patients be resistant to the imatinib, dasatinib or nilotinib. It can also be used for the treatment of adults have not used other TKIs treatment of CML and Ph + ALL. It is the only market drug which is effective in treating the T3151 mutation of the Bcr-Abl kinase Ponatinib has been approved by the EU centralized procedure in July 2013.

Ponatinib, also known as Iclusig, is a small molecule drug used in the treatment of certain types of leukemia. It works by inhibiting the activity of certain proteins called tyrosine kinases that are involved in the growth and survival of cancer cells.

Function:
Ponatinib specifically targets a protein called BCR-ABL, which is created by a genetic abnormality called the Philadelphia chromosome. BCR-ABL is a tyrosine kinase that drives the growth and survival of leukemia cells. By inhibiting the activity of BCR-ABL, ponatinib can stop the growth of leukemia cells and induce their death.

Application:
Ponatinib is primarily used in the treatment of two types of leukemia:

Chronic myeloid leukemia (CML): Ponatinib is used to treat CML that has become resistant to other treatments or has a specific genetic mutation called T315I.

Acute lymphoblastic leukemia (ALL): Ponatinib is used to treat ALL that has a specific genetic mutation called Philadelphia chromosome-positive.
