Glibencla mide, also known as euglycemia and daunorubicin, is a second-generation oral sulfonylurea hypoglycemic agent with the same mechanism of action as toluenesulfonylurea, and its hypoglycemic effect is the strongest among sulfonylureas, with an effect 200-250 times stronger than that of toluenesulfonylurea. It can selectively act on pancreatic beta cells and promote insu lin secretion, enhance the hypoglycemic effect of exogenous insu lin and strengthen the post-receptor effect of insu lin. It is rapidly absorbed orally and has a high protein binding rate. The effect starts in 30 minutes and is strongest in 1.5 hours and lasts for 16 to 24 hours. The volume of distribution is 0.1L/kg, the plasma protein binding rate is 90%-95%, and the half-life is 4-8 hours. It is mainly metabolized by the liver and has six metabolites, two of which are known to be hydroxylates, all of which have no hypoglycaemic effect and are mainly excreted in the urine, with a small amount excreted by the stool. It is mainly used clinically for the treatment of mild and moderate non-insu lin-dependent diabetes mellitus.
