Sorafenib tosylate is a small molecule kinase inhibitor used in the treatment of various cancers, including advanced renal cell carcinoma (RCC), hepatocellular carcinoma (HCC), and differentiated thyroid carcinoma (DTC). It was first approved by the US FDA in 2005 for the treatment of advanced RCC.
The primary function of sorafenib tosylate is to inhibit the activity of multiple tyrosine kinase receptors, including the vascular endothelial gro wth factor receptor (VEGFR), platelet-derived growth fac tor receptor (PDGFR), and RAF kinases. By doing so, it can interfere with the signaling pathways that promote tumor growth and angiogenesis, thus leading to tumor regression.
Sorafenib tosylate has been shown to be effective in extending the progression-free survival and overall survival of patients with advanced RCC and HCC. In addition, it has demonstrated efficacy in other cancers, including melanoma, pancreatic cancer, and non-small cell lung cancer.
The application of sorafenib tosylate is mainly in the treatment of advanced RCC and HCC, where it is often used in combination with other therapeutic agents, such as immunotherapies or chemotherapy. It may also be used in the treatment of DTC that is refractory to radioactive iodine treatment.
